That was my solution, but it was time-consuming. (now I have - I think - over 200 truffle worms)
You need to be in Hardmode for Truffle Worms to spawn in Mushroom Biomes. Also, they're a rare spawn, which means they probably wont be down their everytime you check for one. Also, It *only* spawns underground in naturally made biomes. It will not spawn in surface mushroom biomes or ones you created yourself.

Actually, truffle worms do spawn in artificial mushroom biomes, as long as they are deep enough. You can just make a long, flat stretch of mushroom grass at the depth where the mushroom biomes naturally appear. Then place platforms above it and wait until a truffle worm spawns and then pounce on it, net swinging. Be careful of the other spawns, though. If you don't have at least mid-hardmode equipement, they could be deadly.

And for that matter, if you don't have near end-game equipement, trying to fight the Duke is just pure suicide. But basically, if you could defeat Plantera and the Golem, you should be ready for the Duke.

Um, my artificial underground Truffle Worm farm begs to differ. As long as you're in the "gray background" underground layer, and are in a mushroom biome, the worm CAN spawn. A Water Candle will help with the spawn rates.

Edit: Ninja'd!

I'm arriving to the pumpkin moon to wave 12 without any traps. I've got full spectre and full shroomite set and some pretty good weapons.
