That was my solution, but it was time-consuming. (now I have - I think - over 200 truffle worms)
Actually, truffle worms do spawn in artificial mushroom biomes, as long as they are deep enough. You can just make a long, flat stretch of mushroom grass at the depth where the mushroom biomes naturally appear. Then place platforms above it and wait until a truffle worm spawns and then pounce on it, net swinging. Be careful of the other spawns, though. If you don't have at least mid-hardmode equipement, they could be deadly.
And for that matter, if you don't have near end-game equipement, trying to fight the Duke is just pure suicide. But basically, if you could defeat Plantera and the Golem, you should be ready for the Duke.
Um, my artificial underground Truffle Worm farm begs to differ. As long as you're in the "gray background" underground layer, and are in a mushroom biome, the worm CAN spawn. A Water Candle will help with the spawn rates.
