No regrades
No potions
No arenas
No weapons with ranged attacks (Death Scythe, Terra Blade)
Only thrown weapons

Some ideas (I'll just give these random names):
- Play as Mediumcore (or Hardcore if you want that epic devastation when you die), a bit obvious, but it does add new challenges if you haven't done it. Lava is to be feared.
- 100HP challenge: Don't use crystal hearts, stay at 100HP the whole game.
- Afraid of Heights: Don't use grappling hooks or wings
- Zombie Warrior: At night time, don't wear armour or accessories, grab a low tier weapon (zombie arm if you have it) and go out to battle the zombies (Pre-hardmode)
- Archer: Craft a bow right at the start and only use bows, maybe have a low level backup sword if you run out of arrows
- Mage: Craft a magic staff ASAP, only wear mage robes (diamond robes etc) and only use staffs and other magic weapons
- Melee: Only use non ranged melee weapons
- Potion Junkee: Always drink any potion as soon as you find it or craft it (including health potions). Multiple stacks must be drunk as soon as the original effect runs out.
- Spaceman: Only use Meteor Amour, spacegun, lasergun and any other space-like weapons you can find. Mostly for hardmode.
- Ball on a Chain: Only use flails (once you get your first one) for the rest of the game.
- Trigger Happy: Only use guns (once you get your first one) for the rest of the game. Maybe a low tier sword if you run out of ammo.
- Afraid of Teleports: Don't use the magic mirror or any other teleportation device.

Some I mentioned are included here, as well as heaps more:

The No-Wood challenge. I'm doing two of those right now - one where you can only use cactus and another where you can only use pumpkin - and I have suggested this to other people, too. Initially they never think it is possible but after thinking about it a bit they agree that it certainly is doable. To add extra challenge, restrict your building material to one specific and potentially rare resource like cactus and pumpkin.

Also, what the heck is a "merlock run?" Is that like the Dwarf Run and the Dark Elf Run?

Aren't there things like the furnace and loom that require wood to craft? The furnace in particular would be tough to live without if you were starting out on a fresh world. How does that work? Do you get some cactus armour and try to raid the underworld for a hellforge?

Also another one is make a fresh new world, go kill the world of flesh without building anything, and start a new character (mediumcore) is an untouched hardmode world.

Furnaces do require torches which do require wood in order to make, but remember you can also find torches in pots and chests. You should be able to find enough torches to make a furnace without chopping down any trees.

The Sawmill is a bit trickier; in my first No-Wood Challenge, I found some underground cabins and picked some of the wood in them apart. And I think you can find Sawmills, or Looms, or both, in some underground cabins as well, all ready to go.

I haven't gotten a Sawmille yet in my second No-Wood Chalenge so I don't know what I am going to do about that.

Oh, and cactus armor is WAY too weak for the Underworld. You could still get away with it but it would be rather difficult.

Thanks for all the replys i like the idea of the no-wood.

The Merlock run is were you start with the neptunes shell and a fish bow on your armor slot so you can only be out of water for a certain time and you cant wear full sets of armor.

On topic, maybe a necro armour playthrough?
