Why is this invalid housing?

If I remove the anchor, it becomes valid housing again.

If I place anything else there in that spot (like a different trophy) it becomes invalid.

I'm confused.

for some reason having all the trophies seems to force both flags into the same spot which won't let you put a second NPC in there
this is strange since npcs don't really care about having space for their flag normally, but if you have both they seem to suddenly care a lot
get rid of the anchor, put both NPCs you want inside, then place the anchor back down, OR just have one NPC in there

Does the door work correct while some trophy in place? In other words I suggest you make it one block longer horizontally, the house. And place that anchor one block further from the door. That's just a wild guess though.

Another possibility - not enough room for the NPC's flag on the wall.

Yes. Though in my experience a door doesn't need to work for a house to be valid, as I have blocked the doors with dirt during blood moons and it didn't make invalid housing.

Nah, not going to renovate when I can just remove the decoration.

That shouldn't matter either.. as that is not a physical object and seems to work just fine on all the other various objects and chests I have in the other houses.

Also, I tried putting the anchor in the empty house to the right of that one, in the exact same position relative to the door, and it was a valid house.
Maybe your missing a wall somewhere? Behind the chest maybe?

All the walls are fine. It's something to do with that trophy being in that spot but I don't know why (also tried with eye of cthulu trophy, same thing happened).
it's the slime

Good catch... moving the slime out fixed it. It's weird that it does that but at least there is a workaround.
