Pre-hardmode you can isolate areas with 4+ wide gaps, breaking up the world into chunks between natural caves, but even this is mostly a lost cause once you start breaking altars. Gaps won't prevent the initial diagonal stripes of infection caused by killing the WoF, nor will it stop larger areas from possibly having corruption/hollow added to them when an altar breaks. It can however be used to separate the areas near spawn so that they remain unchanged.
The diagonal stripes generate at the surface about every 1/3 of the map in a v-like shape. This is why vertical shafts usually don't accomplish much. It can be more useful to just separate the upper portion of the terrain below spawn from everything else to give yourself a wedge of safe land to build on, and either use purification powder or a large amount of explosives to create large breaks between where the jungle is (those areas that are entrely mud) and where the rest of the world is. Then just accept that everything else is going to eventually be converted one way or another.
Corruption/hollow won't spread to background.
5 blocks wide is fine as well... Just need to go over it twice to make sure there aren't any missed areas along diagonals where the distances can be shorter. If you're wanting to be extra cautious, you could build 5x5 blocks of non-corrupting material at the corners so that grass cannot spread down the walls to infect other blocks... But this measure is mostly just delaying things.
Corruption only spreads through grass, sand, ice, and stone, while slowly converting mud to dirt then spreading grass to it. Often, what people miss are those small 1 tile bits of corrupted grass near the surface which can allow corruption to jump multiple blocks quickly through dirt but be deep enough that purification powder doesn't reach.
