I’m pretty sure you can get the Biome Keys as soon as you enter Hardmode, but you can’t open the chests until you beat Plantera.

Yeah, and btw try to farm for all the keys, they are all pretty useful in their own way.

All biome keys can be acquired post-WoF, if I'm not mistaken. If not that, then post-Mech bosses. The chests, however, cannot be opened until you defeat Plantera.
