How to use Set Bonus

For example, I have a crimson armor set. To gain addition health regen do I need to hold down S key? Or this bonus will work by itself?

That buff is always on by itself.

Some buffs are passive and other are active.
The few actives that actually require Key input (from the top of my mind).
Chlorophyte (?)
Shroomite (?)
Stardust

Vortex and Forbidden are the only two armor sets I can think of that need activation of their set bonus. For every other set the bonus is always active.

Vortex needs activation because the special stealth bonus slows down the player and Forbidden places a special sentry at a chosen location.

I don't think Stardust works this way anymore. The Guardian engages enemies by itself now, but I haven't been able to send him directly somewhere.

Shroomite was always "Stand still to get stealth and bonus damage".
