Depends on how big your world is.
Also it is really hard to say because you need a WorldEditor to see how deep it really goes.

You need to go all the way to the Underworld. Once Hardmode activates, the Corruption will spread so much faster, and it will go all the way to the bottom. It doesn't spread through ash, though.

use a compass, it shows how far underground you are, on large worlds you have to travel over 200 blocks down from the surface to enter the cavern/underground layer

A signal that your in the underground corruption is that the music changes, although it's more accurate to use a depth meter

Underground is anything below 0 ft. The Caverns layer starts anywhere between 480-960 ft. below that. The cavern layer will have a gray background behind block walls, and will be made mostly of stone, not dirt.
