So far as I can see about the only use for locking a door is to keep a small supply of barley and berries protected from use so you have some available to plant at the start of spring, locking a shelf of each away in a special 3x1 room will stop any settlers getting to it until you unlock the door again. Other than that there doesn't seem to be a use right now, doors in normal usage function as locked for raiders even though they aren't locked, and nothing else potentially needs to be protected from your own settlers in such a manner.
