The game supports local co-op using either keyboard/mouse and one controller, or two controllers.
If you have just one controller plugged in and start a two player game, the keyboard and mouse will be assigned to player 1 and the controller will be assigned to player 2.
If you have two controllers plugged in and start a two player game, then a controller will be assigned to each player, but player 1 can still use the keyboard and mouse if they wish.
In the first scenario (two player with one controller) there is a bug such that when in game, the controller configuration option for player 2 does not appear on the options menu, which may make it difficult to use a controller that needs configuring. This is fixed in the upcoming patch.
Hope this helps :)
