Eve. Doesn't start with good items, has really low damage. Still the best character because of the challenge.
Judas. Highest base damage, highest damage potential, and very rewarding if you can get past his low starting health.
Isaac once you unlock the D6. Without it, Cain.
Between Isaac (When holds the D6) or Cain. Good pills are a great boost in earlier levels. D6 can bring Mom's Knife if you are lucky enough. Eve is a good challenge...
My favorite is isaac, he has the D6 to help him get more health, or better items, than the cookbook or something, he is all around also. My 2nd is judas, high damage and starts with some money, 3rd is cain, he has decent speed and damage and his luck foot helps him get the fly love ability
I actually really liked Magdalene. Thats who I got my first win with her. She's useful as you can get Devil items without taking too much a hit
I think Cain is the best. Luck Foot can really snowball you hard. Isaac is 2nd maybe, but I haven't played him much since I unlocked D6 (trying to get all the achievements for everyone else).

I really like Eve also. I'd call her 3rd best. ♥♥♥♥♥ of Babylon is a really good item, and devil room items can get you 100% demon form. (especially if you get Guppy's Paw, game over, you win).
Maggy + Nun's Habit + poison touch = kill everything with infinite health.

Not really. It only helps in the early floors (before damage gets increased to full hearts) where it's not that needed because the floors are easier. Maggie (Yum <3) + Habit is still great late game, but it's a huge fall off from early game.

I don't see why so many people think that if you have Yum Heart or Book of Revelations with Habit you are invincible, because once you get past mom, that's certainly not the case. That can remain true if you get the Wafer, but all those items are rare enough and very hard to have in one playthrough.

I know that it's not technically invincible, but it makes the first few floors less of a grind if you get Habit early on. The best advantage is being able to take everything from all curse and trap rooms without care and make more deals with the Devil than other characters. Not only that, you can really abuse the blood bank. With the Habit on the first floor with Maggy, you are guaranteed a win. With battery or 9-volt, you are almost guaranteed the win.

Even without a spacebar charge item, you should be able to get past the first 2 floors without getting hit and only taking damage from the curse rooms. By then, you would probably have picked up a speed upgrade. But without the spacebar charge items, I'd put Isaac 1st, since the start bomb and D6 are very useful.

1. Maggy with Habit, Battery, or 9-volt == winning while playing stupidly.
2. Isaac-- D6 to get a whole bunch of good passives early on, then get rid of it if you find a really good spacebar item after beating Mom.
3. Maggy without good luck
4. Cain-- weaker than other characters when you get technology 2 or chemical peel. I just basically use him as Isaac starting with 1 heart and bomb traded for PHD and a tiny bit of starting damage.
5. Eve-- can abuse dead bird and ♥♥♥♥♥ of Babylon while essentially getting a free curse room on the first 2 floors. Sucks when you accidentally make a bad move on the 2nd boss when you don't have spirit hearts.
6. Judas-- Starting with Belial is nice, but I just don't really use it, so to me he's just Cain with worse luck and less health.
7. Blue baby-- unplayable if you don't come across spirit hearts. Only the nail makes him close to playable. Most of the time, he just feels like Isaac without D6 and a huge health disadvantage.
