Did you try to reinstall Skyrim? (clean the whole Skyrim folder from steamapps->common)
Also after you upgrade to windows 10 you need to manually start up the automatic updates because it doesnt start by itself(but only needed to do that the first time).
so:
a. Click Start and type cmd in start search.
b. Right click on Command Prompt to select "Run as Administrator".
c. Type: sfc<space>/scannow.
Note: SFC scan may take a while to complete.
If the issue persists you can try reregistering kernelbase.dll and check.
a. Click Start and type cmd in start search. Right click on Command Prompt and select Run as Administrator.
b. Type "regsvr32 KERNELBASE.dll" without the quotes and Press Enter.
c. Restart your computer and check.
Win10 Right click on the start button, choose "Command Prompt (Admin)"

So, Dependency Walker is your friend.
It may help me that after installing my OS I always run the sfc /scannow, just like after installing a Steam game I run verify game cache. Old school, I prefer a hard copy because I do not trust downloads!!
Make sure that "KERNELBASE.dll" is a valid DLL or OCX file and then try again

You are not going to like MS solution: Copy everything over to the new profile... a &^&^%^& nightmare.
Apparently many others issues can bork KernelBase.dll including bad video drivers/hw, malware/virus. Some programs actually replace that file. So you might want to rename it, back it up too while you are at it, then delete it, and then run sfc /scannow which should d/l a new file.
