am i the only one that has tried to... butcher it for meat and leather?
Ah see I took local leader by level 3 anticipating Fallout sim city, so my perception of what does what in that regard is slightly skewed.
I created a Steam Guide about many topics related to settlements. Whether you are starting your first settlement or are trying to figure out how to maximize your gain from your settlements this guide can be helpful to you. I would love some feedback!

wut
You just highlight the Settler and press the C key and then choose which Settlement he/she should trade between... I don't think the Brahmin is necessary at all to the supply lines.
ok i watched video's for 2 days about settlements while i waited for fallout 4 to download.
and your wrong all you need is the perk once you have it you point at the person and press q to assign their job as a supply line then select the location.
I just now got my first brahmen to show up and ive allready got 3-4 supply lines going, to my other settlements.
You DO need local leader 1 to establish supply lines, it says it in the description of the perk. You DO NOT need the perk to move people between settlements.
You DO NOT need a brahmin to establish a supply line, the ones with saddle bags just spawn when you assign someone to a trade route.
Having a brahmin at your settlement generates fertilizer in the junk section of the work shop. To my knowledge brahmin simply appear once you have people, food, and water, though it does not seem like you need more than 5ish people for one to show up, if that.
Killing them really pisses everyone off, but you can get away with it if no one sees you, or at least I'm assuming, I blasted a person in the face when he was all alone and no one said sh¡t.
Hey everybody I'm a man named Butcher Pete and I'm chopping up all the Brahmins meat.
