You can skip challenges.
So far I'ld say that the tier get more difficult though, so you gotta be more careful on the higher tiers.
The Fracture is endless. Not finishing challenges won't stop you from going into deeper levels. You can always go back to earlier levels to retry the challenges.

- Getting 5 challenges in different floors didn't add up and did not reward me the related achievement. I finally got it in one of the four other dungeons.
- it is said to be rewarded in a randomly generated dungeon and I presume Fracture's levels are the same everyday, not daily randomly generated.
Did you get both achievements at the same time in this way?
