If it's wireless - Check your batteries, (they do not last for ever.) My mouse especially will drain some what fast (and it IS time for me to get some new batteries for it) - Keyboard not so much.
If also it's wireless - do you have other wireless activity going on? (Yes, even WiFi - Cell Phones - Music Player - TV - Tablet, to name a few.)
If it is PS2 interface - the take a can of air and (TURN OFF THE COMPUTER WHEN DOING THIS) - (TURN OF THE COMPUTER WHEN DOING THIS!!) then unplug them from the computer and blow out both connectors - keyboard / mouse and computer connectors ... do not let them "Freeze" though. Do it quickly in very very short bursts.
