"Nobody's going to space, mate!"
I heard this line the first time I played the game but never since then. I remember it was at some point after you attach the Space Core to Wheatley, but not when exactly.
What triggers it exactly?

After you attach the space core, he says something about going to space, then Wheatley replies with this quote.

I've replayed the game a few times but I've never heard that line again. Is it random or am I supposed to wait a bit before hitting him again?

It's after the credits, when Wheatley and the space core are moving around space, before returning to the main menu. :)

It's not after the credits, all that is specific. Same things happen every time. I'd have seen it. All they say is things like "I'm in space!" "I know mate".
Plus they're already in space at that time, why would Wheatley say that? And I do remember it being before they go to space.

I hear it first time i played the game too, but never since then. Maybe removed in an update?

It could just be that you managed to move faster this time, so the game didn't get to that part of the script yet before moving on to the next part.
