I personally started off with single player and ran through all the campaigns from start to finish, chronologically. This is to:
1. Orient yourself with the story, settings, and characters.
2. Get (a little) used to the game's controls and mechanics.
3. Give yourself room to make and learn from mistakes (not that you'll be perfect later on).
Jumping into multiplayer right off the bat will mostly likely leave you bewildered as you'll most likely be thrown into the middle of a game. If you decide to do so, here is my advice:
1. Start off on Easy or Normal. Seriously.
2. Make a lobby yourself and don't jump into a campaign unless you've already played it before, start to end.
3. Listen to the more experienced players and stick with the team. If you don't, you might find yourself literally left for dead, at the receiving end of emotional abuse, or kicked.
4. Avoid Versus and Scavenge until you feel confident or obtain a thick enough skin.
That's it. The most important thing is to have fun. Enjoy!

2nd) As a new player coming into this game, you will have to learn game mechanics and controls. I would suggest single player but if you want to play with others, go into Campaign Normal or Easy difficulty to learn the game mechanics, items, weapons, and map layouts. Take note on how special infected attack, that will help you in Versus much later when you are in control of one them, tasked with attacking survivors. Avoid Versus/Scavenge modes for now until you have built enough experience.
Replay the different campaigns with a harder difficulties or try Survival mode. Later, if you want to practice infected, there are Versus Mods like Raymond's and that Anime modded Server (where players only play as special infected only against survivor bots)

WRONG!!!! SO wrong, I could not even tell if that was sarcastic or not. Single player can be useful for those JUST entering the game for the first time. The bots follow you around, protect you and heal you. Allows you to learn the game at Your Own Pace and not have to keep up with others.
Its secondary use is allowing this game to played offline if an internet connection is not available. It prevents the game from being an online only game like TITANFALL.
I'd say start out on multiplayer campaign normal, then advanced after you start to get the hang of it. The multiplayer experience is what makes this game so fun so I'd avoid single player, even if you're just starting out.
Better to get some experience before going online.
