Does the Japanese have hirigana, katakana and kanji? Perhaps more importantly, does it have romaji too?

Yes. Kana, kanji and romaji.

Yes it has all, you can easily switch between them. It also says the words to you as well.

I must have missed this on the store page, but I am also interested in the Japanese version and am wondering just how many vocab words are contained within the game.

judging by achievements there are 420, Just did a small session, it seems like it may work to get some basic basic words down pretty well. I've already learned a few in about 15 mins which is cool. I just hope they do some basic sentence structure as well!

True, but I find it much easier to learn with romaji, helps with the pronunciation. Plus I'm mainly interested in speaking rather than writing, so it's more beneficial to me.

Does anyone know if there's some kind of grammar or sentence structure featured? Or is it just a nice vocabulary-test-game?

From what I gather it's only vocabulary (I may of course be wrong though).

Learn it in Romaji as long as you want and don't listen to ignorant loud-mouths. If you want to learn Kana or a few Kanji, there are tons of free apps for Android. JA Sensei for example. You can draw them directly with the touchscreen. Pretty neat.

You won't learn Japanese well by using Romaji, it doesn't activate the learning process like using Japanese characters can.
