You'll absolutely need the 4 Seals (Origin, Birth, Life, Death), and have visited/scanned all of the grail tablet locations.
Without knowing more about what you know, or your skill level in navigating the ruins (If you're using a guide, or even if you are, how long it takes to make sense of the matieral, or making a lot of trips to the surface to heal), it's hard to say; but 3 hours is probably a reasonable amount time to perform the tasks you have left. I wouldn't sneak in Hell Temple though if I were you.

Also, what does using the Lamp of Time in Tower of Ruin unlock? I completely forgot about that.
When you use lamp of time in certain room in tower of ruin, you can kill invisible enemies. Then a dais appears and putting wieght to it reveals medicine statue.
Man, its all revolving around the Chamber of Birth and unpleasant pain. This is unnerving.
Even if you don't get in under the 10 hour mark this time, if you replay the game you'll remember most of the puzzle solutions and it should be very trivial to hit that goal on a replay.
Technically no, that specific puzzle does not require either of those things. Solving the puzzle that makes the statue produce the appropriate medicine with which to place in the Vessel? Yeah - for that you need the Djed Pillar & have spoken to the appropriate philosopher; also you'll need the Mantra software, which means you need to have killed Tiamat already, as you can't get that software without the magatama jewel.
Now I just gotta see how I fare when I go full tryhard and just play the game without checking for hints or getting stuff like the spaulder or scriptures.
Also, if you're feeling particularly brave/crazy, you could always try Hard Mode or Hell Temple -- good luck with those, fearless challenger!
I actually beat La Mulana my first time on hard because I'm a ♥♥♥♥♥♥♥ idiot.
If you're looking to do speedrun attempts, be sure to check out Traysandor's any% guide in the steam guides section for La-Mulana - It's really comprehensive, informative, and well written! That'll give you an excellent framework for a speedrun attempt.

Thanks for the promotion, hehe. And yeah while my any% guide doesn't really go into the details and specifics about how to solve the various puzzles, it does give you an idea roughly what to do and whne, and has checkpoints and some screenshots added in so that you can track your progress through the game. Along with boss strategies too. Do note though that the 2 hour in game timer limit isn't too particularly flexible, so you won't have a lot of time to grab extra things. Also Hidden Distance does his runs of the game slightly different than the guide, though it's still mostly the same overall.
