Why?
Media players normally use OpenGL or Direct X as their render and therefore can conflict with the game.
But it is possible to run a full screen game and watch a video on the other monitor (most of the time).
I have VLC running maximized (not fullscreen) on my second monitor while playing ex. Borderlands 2 on my primary monitor in full screen. I don't Alt+Tab since that causes problems with the video, so I use a VLC remote on my Android tablet or iPhone for control or load a video.
