is this achievement over-time or in 1 game?

One play-through. I had trouble with this initially, but if you are looking for a strategy solely for this achievement, I'd recommend taking Flame magic and going into rooms that have 2 horizontal lines of the floor worms / spikes and casting Flame. Each of those count as an enemy, so it's generally much more efficient using in rooms such as those. Hope that helps.

I got it purely by accident on a super long run through hell. Turns out, smart bomb drops count as magic kills, too.

The achievement description on Steam should be fixed then. Other achievs that need to be done in one playthrough tell you. -.-
