Speed helps you stay in front, despite AI rubberbanding so badly that they can outrun Sonic with 6-Speed. You'll stay in front longer with high speed than you would using a character with 1 speed.
Acceleration is important for recovery. In S-class, expect to be bombarded with millions of missiles, snowballs, drones, and whatever the hell they find. You want to get your speed back as quick as possible. Anything less than 3 in my opinion is a bad move for S-class
Handling helps evade weapons more than anything. With enough practice it's easy to navigate a course with just 1 handling (part of the reason Console mods are popular for time attack), but having more handling gives you a better means to dodge attacks, such as swarms and blowfish.
Boost helps more if you know the course layout and are familar with drift and stunt mechanics enough to know where to get the most of them. Like Speed though, S-class racers don't follow the rules, and can still outrun you if you have a 6 in boost and roll a level 6 boost, hell they can outrun All-Star powerups if they're feeling up to it.
All-Star, this is preference. The stat has no real bearing, outside of how powerful the attack of the All-Star powerup for your specific racer is. This stat doesn't follow the 12-point distribution amng other stats, so don't worry too much about it.
Generally I find that I can handily win S-Class races with a character that has 4 Speed/3 Acceleration/3 Handling/2 Boost, but try out different racers and different stat distributions. Lke NBlast points out, you can win anything with anyone.
