Is every playthrough the same? New Game +?
Is there a new game plus?

Not in the sense of increased difficulty similar to games like torchlight 2.

But there are three reasons that are even better:
1. You can play through it again in oldschool mode(No Map) or a harder difficulty if you had fun.
(Its getting way better at:)
2. You can set your own difficulty by playing in an unconventional way(4 mages) or using unorthodox skill builds.
(The main reason though is:)
3. This game includes a map editor and steam workshop integration so you have basically endless content to browse through and knock yourself out.

edit: (almost forgot:)
4. Toorum(find out for yourself...)

While for some games like Skyrim people prefer to avoid the workshop because of its limitations and instead use the Nexus, I cant tell you if this is also true for Grimrock.
But if you wanna be one the safe side you might wanna check out http://grimrock.nexusmods.com/ as well to weigh your options and maybe find mods that aren't available in one or the other site.

Master Quest works much like a New Game + mode, with increased difficulty, changed puzzles, etc. And you can import your final original party to further their leveling up
