I think Ace42 wants you to make sure that Rocksmith is aware that you want to use a mic. In Rocksmith go to the Main Menu; choose Options; and then there might be an "Audio Options" choice and the Rocksmith Mic is probably there.
If Ace42 means the Windows mic settings--go to your win Desktop; press your 'F1' key; and search for "Open Device Manager"; and Windows will create a link to the Device Manager for you.
At this point I am a little lost because I don't know what kind of mic you have. Look in the Device Manager places labelled, "Sound, video and game controllers" or "Human Interface Devices" or "Mice and other pointing devices".
When you find something that looks like your microphone (or headset, or whatever) double-click on it and a small screen wil pop up. There are 3 tabs here each with a bit of infor about whether or not your mic is working, what kind of driver Windows is using for it, and other details.

EDIT:
I just went in the game, it's under Options, then the first thing you can select should have Microphone settings which will give you the optipn to enable the mic. If I remember right mine was not enabled by default
