Many people start as King or Emperor, and then eventually move on to starting as a duke, and then onto starting as a count. (Count is very challenging, and can end -very- quickly.
Good starting Kingdoms are Denmark (at the earliest possible start date), Poland, or one of the various Spanish Kingdoms like Leon, Castile, or Galacia. (Avoid Aragon or Navarra.. they're somewhat harder for a new player, and Navarra can be extremely challenging to the unskilled... sort of like playing as a duke surrounded by a bunch of kings who can easily unite around each other, ally each other, and screw over anyone that screws with any of them. Not to mention Navarra is a frequent French target)
Regarding killing your wife: There's a lot of ways to do it. It's one of the standard plots you can perform and the game will occasionally encourage you to do it. If your wife ever develops the 'chaste' trait, you need to kill her -immediately- or else she won't produce heirs for you and your game will be over. The easiest, but most risky and expensive way is to pay for assassination on the spot, but most people just start up a plot and invite some people to help.
You have little control over the 'how' of her dying if you 'plot' to kill her. One of your conspirators will offer up some plan after a period of time. Assination generally succeeds via poison, and you'll be presented with the 'chance of success' when you select assasinate before you have to pay money and make the attempt. If you fail, the game will roll a discovery chance to see if it can be connected back to you. If you fail -that- check too, she knows -you- tried to kill her and will hate you. She may even attempt to kill you in response, or one of her family members may declare war on you. Killing your wife can be hard, particularly if she's skilled in intrigue, but it's very possible and occasionally encouraged.

Here is my two second summary of how to get a good start.
Marry good nobles (maybe claimants too), fabricate claims with chancellor, keep vassels happy with marriage and getting them to tutor your kids, build buildings that increase levy size first, change succesion law to Primogeniture and send all your sons other than heir to the church (make them bishops, this should make succession crisis less likely.) Start in Ireland as you will have no big rivals.
Other people might say different but this is the way i got a good start on my first proper game.
