I suggest that you play a little vanilla (maybe until you get to around the end of Act 1). This should introduce you to the mechanics nicely and allow you to appreciate the differences between Vanilla and Synergies.
If you do play Synergies, I would also suggest you play as Salan's Necromancer (included). It is balanced very well for Syn and is much more entertaining in my opinion than the base classes.
Also, shameless self-plug here - I have a classmod up named "The Noxmer Class"(can be found in the Workshop). It is mostly balanced for Synergies in Veteran difficulty but might make things that bit too easy in the early game.
Then GO FOR IT. Durring your first time though you learn 'things'. Do it with a character set up that you like. [[Also you might want to concider how much you have played other games of this type]]
Remember that when you use a mod on a certain character that character will be bound meaning if you remove the mod that you have previously used, you won't be able to play that character anymore. But there are some steps to overcome this.
If you prefer to play alone or you've got a crew you always bring along, it can't hurt. TL2 isn't some masterpiece you can spoil by doing it wrong. You're never going to get an adrenaline rush from this game or pull our your hair deciding how to gem your weapon. It's an okay hack & slash like Fate or Titan Quest. Try not to overthink it.
