Tip for Synergy servers to be more Challenging and more realistic
I would really like that there is an option to put users who die can not respawnear until you go to the next map or checkpoint.

Pretty sure that exists as an option already.
Server operators do have this kind of control, however...

Doing what you propose would likely result in many players simply leaving the server to find one they can play on without this kind of restriction.

Just how long would you be willing to sit there waiting for others to reach a checkpoint or finish a map when you could join another server and actually play?

The mp_respawndelay cvar might be adequate for your wants, you'll need to check it out.
If this doesn't give you the desired results you'll need to find or write a plugin using Sourcemod.
Killer to what I mean is that at the time that all die the map will restart, then when someone dies do not respawnee because

I believe the default setting will restart a map when all players die, look for a cvar like mp_restart I think. Then use the mp_respawndelay cvar to set respawn time. You can find cvars by using the find command in the console:
find mp_ will find and list all cvars that have mp_ in them (multi-player cvars)
find spawn will find all cvars that contain the word spawn
